Physical and Chemical Properties
Amino-2,4,6-triiodoisophthaloyl dichloride appears as a yellow to bright yellow crystalline powder under room temperature. It features high iodine content (about 60.5% by weight), which is the core characteristic for its application in contrast agent synthesis. It is slightly soluble in common organic solvents (such as chloroform, dichloromethane) and insoluble in water. It is sensitive to moisture and prone to hydrolysis in humid environments, generating corresponding carboxylic acid and hydrogen chloride gas, so strict moisture isolation is required during storage and use. Its melting point ranges from 228℃ to 232℃, with good thermal stability within the dry and sealed storage range.

Core Application
It is a critical and irreplaceable intermediate in the industrial synthesis of non-ionic low-osmolar iodinated contrast agents, most typically applied in the production of iopromide (Ultravist). Its amino group and two acyl chloride groups have high reactivity: the acyl chloride groups can undergo condensation reaction with hydroxyl-containing compounds, and the amino group can participate in acylation reaction, which provides the core structural basis for constructing the molecular framework of non-ionic contrast agents, ensuring the final product has excellent X-ray absorption performance and biocompatibility. Besides iopromide, it can also be used for the synthesis of other triiodinated non-ionic contrast agents with similar structural types.
Quality Specifications
- Purity (HPLC): ≥ 98.5% (industrial grade); ≥ 99.0% (pharmaceutical intermediate grade)
- Related impurities: Total impurities ≤ 1.0%; single maximum impurity ≤ 0.2%
- Moisture content: ≤ 0.3% (Karl Fischer method, to avoid hydrolysis)
- Free iodine: ≤ 0.05% (to prevent impact on subsequent reaction selectivity)
- Storage: Store in a cool, dry, well-ventilated warehouse, strictly away from moisture, water and light; package in airtight and moisture-proof containers (such as aluminum foil bags or sealed glass bottles)
- Handling: Operate in a dry, inert gas-protected environment; wear protective equipment (gloves, goggles, protective clothing) to avoid contact with skin and mucous membranes
- Safety: It is corrosive, and contact with skin will cause irritation and burns; hydrolysis produces corrosive HCl gas, which is irritating to respiratory tract; it is flammable when exposed to open fire and high temperature, so it should be kept away from fire sources and oxidants during storage and transportation. It is only for industrial and pharmaceutical intermediate synthesis, not for medical and daily use.
